Abstract Fresh cheese is one of the most consumed products of the dairy industry in México. This study aimed to determine the frequency, antimicrobial resistance, and genetic characteristics of extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli in fresh cheese. From fresh cheese samples, E. coli was isolated and the production of ESBL, the antibiotic resistance patterns, the frequency of resistance genes, and the genetic relationship were analyzed. Sixty ESBL-producing E. coli belonging to phylogenetic groups A, C, and B1 were isolated and were highly resistant to the beta-lactam antibiotics, tetracycline, streptomycin, and trimethoprim-sulfamethoxazole. The blaCTX-M gene was detected in all isolates, either alone or in combination with blaTEM and blaSHV. Similarly, a high frequency of resistance genes tetA, strA, and strB and class 1 integrons were found. According to their ERIC-PCR fingerprints, the E. coli were clustered into eight groups. In conclusion, a high frequency of ESBL-producing, genetically diverse, and multidrug-resistant E. coli was found in fresh cheese. The presence of ESBL-producing E. coli in fresh cheese constitutes a public health issue because these bacteria may be pathogenic or contribute to the dissemination of resistance genes to other pathogenic and non-pathogenic bacteria.

Abstract Strawberry (Fragaria x ananassa) is an exquisite food, which provides health benefits, making it the berry with the highest production and export in Mexico. However, it is highly perishable, susceptible to postharvest damage, mainly by Botrytis cinerea and Rhizopus stolonifer, among others. The use of pesticides in preharvest is the control strategy of these pathogens; however, pesticides are documented to harm human health and the ecosystem, showing the need to explore friendly alternatives. Reagent grade chitosan (QGR) is an innocuous polymer with widely reported antifungal activity, while sodium octanoate (8:0) (OS) also has this property; however, so far it is unknown whether commercial grade chitosan (QGC) (the most suitable candidate to conduct a larger commercial study, due to its cost), available in Mexico, has the same effect. Therefore, the objective of this research was to evaluate the effectiveness of QGR and the QGR-OS composite in the protection of strawberry in postharvest and compare it with that of QGC and that of the QGC-OS composite. The compounds were sprayed on the fruits and incubated simulating export conditions. The results showed significant reduction in the severity and incidence of postharvest strawberry fungal diseases after the application of QGR, QGC and QGR-OS, QGC-OS, but not that of OS applied alone. QGC and the QGC-OS composite are excellent candidates for use in a larger commercial study.

Resumen La fresa (Fragaria x ananassa) es un alimento exquisito, que proporciona beneficios a la salud, por lo que es la frutilla de mayor producción y exportación en México. Sin embargo, es altamente perecedera, susceptible de sufrir daños en poscosecha, por Botrytis cinerea y Rhizopus stolonifer, entre otros. El uso de plaguicidas en precosecha es la estrategia de control de estos patógenos; no obstante, está documentado que los plaguicidas dañan la salud humana y al ecosistema, lo que muestra la necesidad de estudiar alternativas amigables. El quitosano grado reactivo (QGR) es un polímero inocuo con actividad antifúngica ampliamente reportada, mientras que el octanoato de sodio (8:0) (OS) también tiene esta propiedad; sin embargo, hasta el momento se desconoce si el quitosano grado comercial (QGC) disponible en México, tiene el mismo efecto. Por lo tanto, el objetivo de esta investigación fue evaluar la efectividad de QGR y el compósito QGR-OS en la protección de fresa en poscosecha y compararla con la de QGC y el compósito QGC-OS. Los compuestos se asperjaron sobre los frutos y se incubaron simulando las condiciones de exportación. Los resultados mostraron reducción significativa de la severidad y la incidencia de las enfermedades fúngicas de fresa en poscosecha luego de la aplicación de QGR, QGC y QGR-OS, QGC-OS, no así del OS aplicado solo. El QGC y el compósito QGC-OS son excelentes candidatos para ser utilizados en un estudio comercial de mayor alcance.

Abstracts Escherichia coli is an environmental bacterium frequently implicated in bovine mammary infections. Although specific virulence factors are unknown, biofilm-forming bacteria are associated with persistent infections, and motile bacteria exhibit increased virulence. An analysis was done of the genetic relationship, biofilm formation capacity and motility of bovine mastitis-associated E. coli, as well as the in vivo virulence of representative bacterial isolates. Of the 34 isolates, 67.7 % belonged to phylogenetic group A, 17.6 % to group B1 and 14.7 % to group D. Genetic relationship analysis done with (GTG)5-PCR indicated that the analyzed bacteria are diverse, with only two isolates exhibiting 100 % similarity, and the remaining 32 being grouped into seven main clusters with more than 70 % similarity. Biofilm formation capacity ranged from strong to moderate and weak in 76.5 % of the isolates. The csgA and fimA genes were detected in 52.9 % of the biofilm-forming isolates. Most (70.6 %) of the isolates were motile. In vivo infection assays using Galleria mellonella showed the biofilm-forming bacteria to be more pathogenic than the non-biofilm-forming ones. The studied bovine mastitis-associated E. coli were genetically diverse. Biofilm formation capacity and motility were variable among the isolates, but the biofilm-forming bacteria were more pathogenic than the non-biofilm-forming ones.

Resumen Escherichia coli es una de las bacterias ambientales que frecuentemente se ha visto implicada en infecciones de la glándula mamaria bovina. Aunque se desconocen los factores de virulencia específicos involucrados en dicho proceso, se ha observado que las bacterias formadoras de biopelículas se asocian con infecciones persistentes. De igual forma se ha planteado que las bacterias móviles pueden ser más patógenas. El propósito del trabajo fue analizar la relación genética, capacidad de formación de biopelículas y movilidad de E. coli aislada de casos de mastitis bovina, así como la virulencia in vivo de aislados representativos. El 67.7 % de las bacterias pertenece al grupo filogenético A, el 17.6 % al grupo B1 y el 14.7 % al D. El estudio de la relación genética mediante (GTG)5 reveló que solo dos bacterias son idénticas genéticamente, el resto se organizaron en siete grupos distintos con 70 % de similitud. El 76.5 % de las bacterias fue capaz de formar biopelículas de forma fuerte, moderada o débil. Además, los genes csgA y fimA se detectaron en el 52.9 % de las bacterias formadoras de biopelículas. El análisis de movilidad mostró que el 70.6 % fue móvil. Mientras que, el análisis de patogenicidad en larvas de Galleria mellonella reveló que bacterias formadoras de películas y móviles fueron capaces de matar un mayor número de larvas a las 24 h que las no formadoras. Los resultados indican que las E. coli causantes de mastitis bovina son muy diversas y pertenecen principalmente al grupo filogenético A y que las formadoras de biopelículas y móviles son más patógenas.

Abstract: This study describes the antibiotic resistance patterns of Staphylococcus aureus isolates from cows with mastitis, as well as their in vitro susceptibility to nisin, chitosan and the nisin/chitosan composite. From milk samples obtained from sick cows, 53 S. aureus isolated were identified, the profile of antibiotic resistance was analyzed. The results showed a high frequency of resistance to beta-lactams group, mainly penicillin, dicloxacillin and ampicillin. In addition, multiresistance was detected in some isolates, since they showed tolerance to 3 or 4 different groups of antibiotics, highlighting the isolates AMC-9 (resistant to three groups) and AMC-23 (resistant to four groups). It is also noted that all isolates were susceptible to levofloxacin. On the other hand, the in vitro susceptibility of the isolates to nisin, chitosan and the nisin/chitosan composite was analyzed determining the Minimum Inhibitory Concentration (MIC). The results indicated 100 % of the isolates were susceptible to chitosan and to the nisin/chitosan composite and 47.1 % to nisin. The effect of chitosan and nisin/chitosan composite on the multi-resistant isolates AMC-9 and AMC-23 was bactericidal. These results show the potential of the antimicrobial compounds chitosan, nisin/chitosan to be used as therapeutic agents against bovine mastitis.

Resumen: En este trabajo se describen los patrones de resistencia a antibióticos de aislados de Staphylococcus aureus obtenidos de vacas con mastitis, así como su susceptibilidad in vitro a nisina, quitosano y al compósito nisina/quitosano. A partir de muestras de leche obtenidas de vacas enfermas se aislaron e identificaron 53 S. aureus, a los cuales se les analizó el perfil de resistencia a antibióticos. Los resultados evidenciaron una alta frecuencia de resistencia hacia el grupo de los beta-lactámicos, principalmente a penicilina, dicloxacilina y ampicilina. Asimismo, se detectó multirresistencia en algunos aislados, ya que mostraron tolerancia a 3 o 4 grupos de antibióticos, destacando los aislados AMC-9 (resistente a tres grupos) y AMC-23 (resistente a cuatro grupos). También se resalta que todos los aislados fueron susceptibles a levofloxacina. Por otra parte, se analizó la susceptibilidad in vitro de los aislados a nisina, quitosano y al compósito nisina/quitosano, mediante la determinación de la concentración mínima inhibitoria. Los resultados indican que el 100 % de los aislados fue susceptible a quitosano y al compósito nisina/quitosano y el 47.1 % a la nisina. El efecto del quitosano y del compósito nisina/quitosano sobre los aislados multirresistentes AMC-9 y AMC-23 fue bactericida. Estos resultados revelan el potencial de los compuestos antimicrobianos quitosano y nisina/quitosano para ser utilizados como agentes terapéuticos contra la mastitis bovina.

Abstract Introduction Grey mold is one of the main causes of post-harvest strawberry (Fragaria x ananassa Duch.) rot fruit, which is caused by Botrytis cinerea Pers.:fr fungus. The treatment with chemical fungicides is the main method for its control; however, its use cause health concerns and pollution. The above promote the research toward alternatives of control. The aim of the present research was to evaluate the chitosan-fatty acids composites effects on the B. cinerea infection in post-harvest strawberry fruits. Method In vitro bioassays were conducted to evaluate the chitosan, sodium octanoate and decanoic acid effect on the mycelial growth of B. cinerea. Likewise, in vivo bioassays were conducted to evaluate the compounds alone and in mixture effect on infection severity of grey mold in post-harvest strawberry fruits. The treatments were evaluated in a completely randomized design and the data were analyzed by means of analysis of variance (ANOVA) (p ≤ 0.05) and average comparison by Tukey´s test (p ≤ 0.05). Results The chitosan inhibited 100 % of the growth when it was applied in 10, 12.5 and 15 mg/mL concentrations, while the sodium octanoate reached the same inhibition percentage in 0.33 and 0.49 mg/mL concentrations. Finally, decanoic acid inhibited in 100 % to B. cinerea in 0.17, 0.34 and 0.51 mg/mL concentrations. In in vivo bioassays, the best results of strawberry fruits protection from B. cinerea infection were those in which the chitosan/sodium octanoate (12.5/0.49 y 15/0.49 mg/mL) were utilized, with absence of severity. Also highlight the chitosan (15 mg/mL), decanoic acid (0.51 mg/mL) and chitosan/decanoic acid composites (12.5/0.51 and 15/0.51 mg/mL) treatments, with severity degrees between 1 and 2. Discussion or Conclusion The chitosan, sodium octanoate and decanoic acid significantly inhibited the in vitro mycelial growth of B. cinerea. The chitosan/sodium octanoate composite have a greater protective effect of strawberry fruits, in relation to the compounds applied alone. The chitosan and decanoic acid presented a significantly protective effect of the fruits, applied alone as well as in composite. The above suggest that these compounds could be potentially used in the post-harvest control of B. cinerea.

Resumen Introducción El moho gris es una de las principales causas de pudrición en poscosecha del fruto de fresa (Fragaria x ananassa Duch.), la cual es ocasionada por el hongo Botrytis cinerea Pers.:fr. El tratamiento con fungicidas químicos es el método principal de su control; no obstante, su uso ocasiona problemas de salud y contaminación. Lo anterior promueve la investigación hacia alternativas de control. El objetivo de la presente investigación fue evaluar el efecto de compósitos de quitosano-ácidos grasos sobre la infección de B. cinerea en frutos de fresa en poscosecha. Método Se realizaron bioensayos in vitro para evaluar el efecto del quitosano, octanoato de sodio y ácido decanoico sobre el crecimiento micelial de B. cinerea. Asimismo, en bioensayos in vivo se evaluó la protección de los compuestos solos y en mezcla sobre la severidad de la infección del moho gris en frutos de fresa en poscosecha. Los tratamientos se evaluaron bajo un diseño completamente al azar, y los datos se analizaron mediante análisis de varianza (ANOVA) (p ≤ 0.05) y comparación de medias por el método de Tukey (p ≤ 0.05). Resultados El quitosano inhibió el crecimiento en 100 % cuando se aplicó en concentraciones de 10, 12.5 y 15 mg/mL, mientras que el octanoato de sodio alcanzó el mismo porcentaje de inhibición en las concentraciones de 0.33 y 0.49 mg/mL. Finalmente, el ácido decanoico inhibió en 100 % a B. cinerea en las concentraciones de 0.17, 0.34 y 0.51 mg/mL. En los bioensayos in vivo, los mejores resultados de protección de los frutos de fresa de la infección por B. cinerea fueron aquellos en los que se utilizó el compósito quitosano/octanoato de sodio (12.5/0.49 y 15/0.49 mg/mL) con ausencia de severidad. También destacaron los tratamientos de quitosano (15 mg/mL), ácido decanoico (0.51 mg/mL) y del compósito quitosano/ácido decanoico (12.5/0.51 y 15/0.51 mg/mL), donde el grado de severidad fue entre 1 y 2. Discusión o Conclusión El quitosano, octanoato de sodio y ácido decanoico inhibieron significativamente el crecimiento micelial in vitro de B. cinerea. El compósito quitosano/octanoato de sodio posee un efecto protector mayor de los frutos de fresa, en relación a los compuestos aplicados solos. El quitosano y el ácido decanoico presentaron un efecto protector significativo de los frutos, aplicados solos o como compósito. Lo anterior sugiere que estos compuestos podrían ser utilizados potencialmente en el control de B. cinerea en poscosecha.

ABSTRACT: In the present study the antibiotic resistance patterns of E. coli isolated from bovine mastitis, as well as the presence of Extended-spectrum β-lactamases (ESBL), tetracycline, streptomycin, and quinolone resistance genes were analyzed. From mastitic cows, milk samples were obtained, of which E. coli were isolated and identified biochemically and confirmed at the molecular level. Genetic analysis revealed that 11.8 % of the E. coli contained a virulence gene encoding intimin (eaeA). All E. coli were resistant to one or more antibiotics, with higher rates of resistance to tetracycline, ampicillin, and cephalothin. In addition, 73.5 % of E. coli were resistant to three or more antibiotics. Of eleven resistance genes analyzed, seven were detected in 79.4 % of the bacterial isolates. Of these, bla CTX-M was found in 16 isolates, tetB in 11, tetA, strA and strB in nine, qnrB in four, and bla TEM in one isolate. About 5.9 % of the E. coli isolates carried class 1 integron integrase gene. In conclusion, a high prevalence of antibiotic resistance in E. coli isolated from bovine mastitis was identified. The bacteria also harbor a virulence gene related to human pathogens and genes conferring resistance to beta-lactam antibiotics, tetracycline, streptomycin, and quinolones.

RESUMEN: En el presente estudio se describen los patrones de resistencia a antibióticos de E. coli aislada de mastitis bovina, así como la presencia de genes de virulencia y de resistencia. A partir muestras de leche obtenidas de vacas con mastitis se aislaron e identificaron a nivel bioquímico y molecular cepas de E. coli, a las cuales se les analizó el perfil de resistencia a antibióticos y mediante PCR se investigó la presencia de genes de virulencia, de resistencia a antibióticos beta-lactámicos, tetraciclina, estreptomicina y quinolonas; así como la presencia de integrones clase 1. El análisis genético reveló que 11.8 % de los aislados de E. coli presentaron el gen de virulencia que codifica para la intimina (eaeA). Por otro lado, todas las E. coli fueron resistentes a uno o más antibióticos, con alta frecuencia de resistencia para tetraciclina, ampicilina y cefalotina. Además, el 73.5 % fueron resistentes a tres o más antibióticos. De 11 genes de resistencia analizados, se confirmó la presencia de siete, distribuidos en el 79.4 % de los aislados bacterianos. De los cuales, bla CTX-M se encontró en 16 aislados, tetB en 11, tetA, strA y strB en 9, qnrB en cuatro y bla TEM en un solo aislado. También, en el 5.9 % de los aislados de E. coli se identificó la presencia de integrones clase 1. En conclusión, se encontraron elevados índices de resistencia en las E. coli aisladas de mastitis bovina. Estas bacterias contienen genes de virulencia relacionados a patógenos de humano; también llevan genes de resistencia a beta-lactámicos, tetraciclina, estreptomicina y quinolonas.

Abstract Seventy percent of water is used in agricultural activities worldwide. However, due to the uncontrolled growth of the population and the contamination of superficial fresh water bodies the water scarcity has augmented, for this the availability of good quality water for agriculture is limited. One of the main pollutants is antibiotic resistant and potentially pathogenic bacteria. P. aeruginosa is a bacterium that normally is found in water and soil, it is pathogen from animals and plants. This bacterium has been considered a resistance phenomenon due to the diversity of mechanisms that contain which represent a potential risk to the population. Method: Samples of agricultural usage water were collected, of which P. aeruginosa was isolates and identified at the biochemical and molecular level. Also, the distribution of virulence genes was analyzed by PCR. The resistance to 20 antibiotics was tested by disk diffusion and microdilution assays. Even more, heavy metal resistance was analyzed by microdilution assays. Results: Forty-six P. aeruginosa were identified at molecular and biochemical level, the bacteria were potentially pathogenic because the following virulence genes was detected, algD and toxA in100% of bacteria, lasB in 97.8%, plcH in 95.6%, plcN in 91,3% and exoS in 89.1%. High rates of resistance were detected in bacterial isolates for ampicillin, ceftriaxone, chlroramphenicol, cephalotin, cefotaxime, nitrofurantoin, kanamycin, streptomycin and tetracycline. Similarly, susceptibility was detected mainly to ceftazidime, gentamycin, imipenem, tecarcillin, aztreonam, levofloxacin, netilmicin and carbenicillin. All bacteria were multirresistant as they tolerate between 8 and 11 antibiotics. According to the observed resistance patterns bacteria were classified in 11 groups. Whereas, metal tolerance was observed mainly for Cu+2, Zn+2, Ba+2, Pb+2 y Se+4. Conclusion: P. aeruginosa isolated from agricultural usage water samples in the studied zone are potentially pathogenic due to the presence of virulence genes. Even more, high rates of antibiotic and heavy metal resistance were observed.

Resumen El 70% del agua a nivel mundial se destina principalmente para las actividades agrícolas. Sin embargo, debido al crecimiento descontrolado de los asentamientos urbanos se ha acentuado la escasez y contaminación de los cuerpos de agua superficiales, de tal forma que cada vez este líquido de buena calidad es más escaso para la agricultura. Uno de los principales contaminantes son las bacterias potencialmente patógenas resistentes a los antibióticos. Pseudomonas aeruginosa es una bacteria que se encuentra normalmente en el agua y suelo, es patógeno de animales y plantas. Esta bacteria se ha considerado un fenómeno de resistencia debido a la diversidad de mecanismos que posee, por lo cual representa un riesgo potencial para la población. Método: Se colectaron muestras de agua de uso agrícola, a partir de las cuales se aislaron e identificaron a nivel bioquímico y molecular P. aeruginosa. En estas bacterias se determinó la distribución de 6 genes de virulencia por PCR. Mediante ensayos de difusión en disco y microdilución se analizó la resistencia a 20 antibióticos. Además mediante ensayos de microdilución se determinó la resistencia a metales pesados. Resultados: Se identificaron a nivel bioquímico y molecular 46 aislados de P. aeruginosa, éstas son potencialmente patógenas ya que en ellas se detectó la presencia de los genes de virulencia algD y toxA (100%), lasB (97.8%), plcH (95.6%), plcN (91.3%) y exoS (89.1%). Dichas bacterias presentaron altos índices de resistencia a ampicilina, ceftriaxona, cloranfenicol, cefalotina, cefotaxima, nitrofurantoína, kanamicina, estreptomicina y tetraciclina. De igual forma fueron muy susceptibles a ceftazidima, gentamicina, imipenem, ticarcilina, aztreonam, levofloxacina, netilmicina y carbenicilina. Todas las bacterias fueron multirresistentes ya que toleraron entre 8 y 11 antibióticos. De acuerdo a los patrones de resistencia observados, las bacterias se clasificaron en 11 grupos. Mientras que para los metales pesados se presentaron altos niveles de tolerancia para Cu+2, Zn+2, Ba+2, Pb+2 y Se+4. Conclusión: Las P. aeruginosa aisladas de agua de uso agrícola en la zona de estudio son potencialmente patógenas debido a la presencia de genes de virulencia. Además presentaron altos índices de resistencia a antibióticos y tolerancia metales pesados.
